Wa from the previous lesson is a word for this.
As for saying no
Desu means it is. When answering and is used when your saying yes to the question. When saying no you would replace desu with ja arimasen.
Example
Hai, kore wa pen desu.
Iie, kore wa pen ja arimasen
Is this a pen or a book?
Kore wa pen desu ka hon desu ka.
When using desu ka. You put it after the two words you are asking about.(Pen=pen/hon=book)
Not either: demo arimasen
Kore wa pen desu ka? Iie, kore wa pen ja arimasen.
Is this a pen? No this is not a pen.
Kore wa hon desu ka? Iie, kore wa hon demo arimasen.
Is this a book? No this is not a book either.
